Western blot analysis of Lane 1: human peripheral blood monocyte (untreated) and Lane 2: human peripheral blood monocyte (treated with PMA/lonomycin) with IL18 recombinant monoclonal antibody, clone SAIC-24B-4 (Cat # RAB03572).
Western Blot analysis the lysates (35ug protein in RIPA buffer) were resolved on a 15% SDS PAGE gel and blots were probed with the chimeric r version of RAB03572 at 0.3 ug/mL before detection using an anti-rondary antibody. Protein was detected by chemiluminescence.

Flow cytometric analysis of human peripheral blood monocytes with IL18 recombinant monoclonal antibody, clone SAIC-24B-4 (Cat # RAB03572).
The human peripheral blood monocytes were fixed using 2% PFA and permeabilized 0.5% Triton and stained with unimmunized r antibody antibody (isotype control-black line) or the chimeric r version of RAB03572 (blue line) at a dilution of 1:100 for 1h at RT. An additional population of PBMC's stimulated with PMA and Ionomycin in the presence of Monensin for 4 hours were stained in a similar manner (purple line). After washing the bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
