Western Blot (Cell lysate)
Western blot analysis of NIH3T3 (A), Raw264.7 (B), Hela (C), A549 (D), HL60 (E), U2OS (F), C6 (G) whole cell lysates with NAMPT recombinant monoclonal antibody (Cat # RAB02618).
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)
Immunohistochemical analysis of human lung cancer formalin fixed paraffin embedded tissue section using NAMPT recombinant monoclonal antibody (Cat # RAB02618). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.101). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescence
Immunofluorescent analysis of U87MG cells with NAMPT recombinant monoclonal antibody (Cat # RAB02618). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.

This gene encodes a protein that catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, one step in the biosynthesis of nicotinamide adenine dinucleotide. The protein is an adipokine that is localized to the bloodstream and has various functions, including the promotion of vascular smooth muscle cell maturation and inhibition of neutrophil apoptosis. It also activates insulin receptor and has insulin-mimetic effects, lowering blood glucose and improving insulin sensitivity. The protein is highly expressed in visceral fat and serum levels of the protein correlate with obesity. This gene has a pseudogene on chromosome 10. [provided by RefSeq
