Immunofluorescent staining of U937 cells with CCR5 (PKC site) recombinant monoclonal antibody, clone RC-10 (Cat # RAB03135).
Immunofluorescence analysis of paraformaldehyde fixed U937 cells on Shi-fix™ coverslips stained with the chimeric r version of RAB03135 at 10 ug/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 ug/ml)- showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right- top-bottom RAB03135- DAPI- merged channels and an isotype control. The isotype control was an unknown specificity antibody (.1) followed by staining with Alexa Fluor® 488 secondary antibody.

Flow Cytometry

Flow cytometric analysis of human peripheral blood monocytes with CCR5 (PKC site) recombinant monoclonal antibody, clone RC-10 (Cat # RAB03135).
Paraformaldehyde fixed human peripheral blood monocytes permewith 0.5% Triton were stained with anti-unknown specificity antibody (3.0; isotype control- black line) or the r version of RAB03135 (blue line) at a dilution of 1:100 for 1h at RT. After washing- the bound antibody was detected using a goat anti-r AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Gene Summary

This gene encodes a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein-coupled receptors. This protein is expressed by T cells and macrophages, and is known to be an important co-receptor for macrophage-tropic virus, including HIV, to enter host cells. Defective alleles of this gene have been associated with the HIV infection resistance. The ligands of this receptor include monocyte chemoattractant protein 2 (MCP-2), macrophage inflammatory protein 1 alpha (MIP-1 alpha), macrophage inflammatory protein 1 beta (MIP-1 beta) and regulated on activation normal T expressed and secreted protein (RANTES). Expression of this gene was also detected in a promyeloblastic cell line, suggesting that this protein may play a role in granulocyte lineage proliferation and differentiation. This gene is located at the chemokine receptor gene cluster region. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq
