Western Blot analysis were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of 304 before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.Immunofluorescence
Immunofluorescence analysi of paraformaldehyde fixed Caco-2 cells stained with the chimeric rabbit IgG version of 304 (1:1000 dilution) for 1h followed by Alexa Fluor 488 secondary antibody (1:1500 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom, DAPI, merged channels and an isotype control. The isotype control was anti-fluorescein antibody followed by staining with Alexa Fluor 488 secondary antibody.Flow Cytometry
Flow-cytometry were stained with an isotype control (black line) or the rabbit-chimeric version of 304 (blue line) at a concentration of 1 ug/mL for 30 mins at RT. After washing, bound antibody was detected using Alexa Fluor 488 conjugated goat anti-rabbit IgG and cells analysed on a FACSCanto flow-cytometer. -

The protein encoded by this gene is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces an apoptosis signal. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. Two transcript variants encoding different isoforms and one non-coding transcript have been found for this gene. [provided by RefSeq
