Western blot was performed on nuclear extracts from KAS-6/1 cells (human myeloma cell line, lane 1) and SKNO-1 cells (human acute myeloblastic leukaemia, lane 2) with RUNX1T1 polyclonal antibody (Cat # PAB14127), diluted 1 : 1,000 in TBS-Tween containing 5% skimmed milk. The location of RUNX1T1 and a presumed splice variant (missing 106 C-terminal amino acids) are indicated on the right.
Western Blot (Recombinant protein)
Western blot was performed on seven GST-fusion proteins containing different fragments of RUNX1T1 (Lane 1~7). RUNX1T1 polyclonal antibody (Cat # PAB14127) was used at dilution 1 : 1,000 in TBS-Tween containing 5% skimmed milk.
ELISA was performed using a serial dilution of RUNX1T1 polyclonal antibody (Cat # PAB14127), crude serum and flow through in antigen coated wells. The synthetic peptide used to immunise the rabbits, served as antigen. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1 : 5,250.
