Western Blot (Cell lysate)
B. Western blot was performed on whole cell lysates from mouse neural stem cells (NS), transfected with GFP tagged Ash2, with Ash2l polyclonal antibody (Cat # PAB14060), diluted 1 : 500 in BSA/PBS-Tween.
Western Blot (Cell lysate)
Western blot was performed on whole cell lysates from mouse fibroblastst (Lane 1, NIH/3T3) and embryonic stem cells (Lane 2, E14Tg2a) with Ash2l polyclonal antibody (Cat # PAB14060), diluted 1 : 1,000 in BSA/PBS-Tween.
Immunofluorescence
Immunofluorescent staining of NIH/3T3 cells. Cells were fixed with 4% formaldehyde for 10' and blocked with PBS/TX-100 containing 5% normal goat serum and 1% BSA. The cells were immunofluorescently labelled with the Ash2l polyclonal antibody (left) diluted 1:200 in blocking solution followed by an anti-rabbit antibody conjugated to Alexa488. The middle panel shows staining of the nuclei with DAPI. A merge of the two stainings is shown on the right.
Enzyme-linked Immunoabsorbent Assay
ELISA was performed using a serial dilution of Ash2l polyclonal antibody (Cat # PAB14060).
By plotting the absorbance against the antibody dilution, the titer of the crude serum was estimated to be 1 : 24,000.
