Applications
Western Blot (Cell lysate)
Western blot using FANCC polyclonal antibody (Cat # PAB10046) shows detection of a band at ~63 KDa (arrowhead) corresponding to FANCC present in a HeLa whole cell lysate.
The identity of the lower molecular weight band is unknown.
Approximately 35 ug of lysate was separated by 4-20% Tris Glycine SDS-PAGE.
After blocking, the membrane was probed overnight at 4°C with the primary antibody diluted to1:1,500 in PBS supplemented with 1% normal goatserum and 0.1% BLOTTO.
The membrane was washed and reacted with a 1:10,000 dilution ofIRDye™800 conjugated Gt-a-Rabbit IgG [H&L] for 45 min at room temperature (800 nm channel, green).
Molecular weight estimation was made bycomparison to prestained MW markers (indicated at left).
IRDye™800 fluorescence image was captured using the Odyssey® Infrared Imaging System developed by LI-COR.

The Fanconi anemia complementation group (FANC) currently includes FANCA, FANCB, FANCC, FANCD1 (also called BRCA2), FANCD2, FANCE, FANCF, FANCG, FANCI, FANCJ (also called BRIP1), FANCL, FANCM and FANCN (also called PALB2). The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex.
