Immunohistochemistry (Frozen sections)
Confocal microscopy on cytospin - isolated human blood cells. A : Neutrophils seem to be stained. B : The cells were double - stained for ATG7 appearing in red and Myeloperoxidase (MPO) appearing in green. Using rabbit ATG7 polyclonal antibody (Cat # PAB0742) at a dilution of 1 : 100, incubated for 1 hr at room temperature. The cells stained for ATG7 appear in red. The cells were counter stained with Hoechst Dye (blue colour) . Here, the merged picture is presented. Observations : No staining was observed using the Pre - immunisation serum. No staining was observed in HL-60 cell line.
This gene was identified based on homology to Pichia pastoris GSA7 and Saccharomyces cerevisiae APG7. In the yeast, the protein appears to be required for fusion of peroxisomal and vacuolar membranes.
