ChIP
ChIP results obtained with the Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656).
ChIP assays were performed using mouse embryonic stem cells (ES14).
ChIP on chip
ChIP-on-chip profile using the Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656).
The DNA of four ChIP assays was pooled and amplified. Subsequently, the DNA was hybridized on a tiling array.

Western blot analysis using the Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656).
Mouse embryonic stem cells (ES14) cells were treated or not with all-trans retinoic acid (ATRA).
The ATRA treatment is used to induce cell differentiation.
Histone (acid) extracts of treated (lane 1) and non-treated cells (lane 2).
Using Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656) at dilution 1 : 1,000 in TBS-Tween + 5% skimmed milk. The location of the protein of interest is indicated with an arrow.
Enzyme-linked Immunoabsorbent Assay
ELISA is a quantitative method used to determine the concentration of a primary antibody using a series of dilutions of crude sera, affinity purified Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656) and flow-through in antigen coated wells.
The antigen used in this case is the peptide including the histone modification of interest.
We plotted the absorbance versus antibody dilution to estimate the TITER : 1 : 12,700 for crude serum and 1 : 1,600 for Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656).
Dot Blot
Dot Blot is performed to test the cross-reactivity of the affinity purified Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656) with other histones and other histone modifications.
Other histone modifications include mono- and dimethylation of the same lysine and mono-, di- and trimethylation of adjacent lysines.
To determine the cross-reactivity, 0.2 to 100 pmol of peptide containing the respective histone modifications were spotted on a membrane.
The antibody was used at a dilution of 1 : 5,000.
At this dilution, a signal is detected at an amount of peptide as low as 0.2 pmol with the Histone H3 (K36me3) polyclonal antibody (Cat # PAB0656).
