Theoretical MW (kDa):
Baculovirus infected insect cell (Sf21) expression system
Glutathione sepharose chromatography
90 % by SDS-PAGE/CBB staining
The activity was measured by off-chip mobility shift assay. The enzyme was incubated with fluorescence-labeled substrate and Mg (or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by LabChip™3000. Substrate : GS peptide. ATP: 100 µM.
Quality Control Testing:
SDS-PAGE Stained with Coomassie Blue
In 50 mM Tris-HCl, 150 mM NaCl, pH 7.5 (0.05% Brij35, 1 mM DTT, 10% glycerol)
Store at -80°C. Aliquot to avoid repeated freezing and thawing.
The protein encoded by this gene is a member of the serine/threonine kinase family. In response to cellular stress and proinflammatory cytokines, this kinase is activated through its phosphorylation by MAP kinases including MAPK1/ERK, MAPK14/p38-alpha, and MAPK11/p38-beta. In vitro, this kinase phosphorylates heat shock protein HSP27 at its physiologically relevant sites. Two alternately spliced transcript variants of this gene encoding distinct isoforms have been reported. [provided by RefSeq
