Human MAP2K7 (NP_660186.1, 1 a.a. - 419 a.a.) K149M mutant full-length recombinant protein with GST tag expressed in Escherichia coli.
Theoretical MW (kDa):
Escherichia coli expression system
Glutathione sepharose chromatography
55 % by SDS-PAGE/CBB staining
The activity was determined by ELISA. The enzyme was incubated with GST-fused substrate protein, and after stopping kinase reaction by EDTA, the reaction solution was transferred into glutathione- coated plate. Phosphorylation was detected by anti-phospho antibody and HRP-labeled anti-rabbit IgG. Substrate: JNK1. ATP: 10 uM.
Quality Control Testing:
Loading 1 ug protein in SDS-PAGE
In 50 mM Tris-HCl, 150 mM NaCl, pH 7.5 (0.1% CHAPS, 1 mM DTT, 10% glycerol)
Store at -80°C. Aliquot to avoid repeated freezing and thawing.
The protein encoded by this gene is a dual specificity protein kinase that belongs to the MAP kinase kinase family. This kinase specifically activates MAPK8/JNK1 and MAPK9/JNK2, and this kinase itself is phosphorylated and activated by MAP kinase kinase kinases including MAP3K1/MEKK1, MAP3K2/MEKK2,MAP3K3/MEKK5, and MAP4K2/GCK. This kinase is involved in the signal transduction mediating the cell responses to proinflammatory cytokines, and environmental stresses. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found, but only one transcript variant has been supported and defined. [provided by RefSeq
