Serial dilutions of mouse Il21 (starting at 100 ng/mL) or ConA (+ control) were added to primary mouse thymocytes cultured on an anti-cD3e coated plate. After 72 hours, total live thymoctyes were measured and the linear portion of the curve was us used to calculate the ED50.
