Western Blot analysis of JAK2 phosphorylation expression in Jurkat cell lysates treated with Pervanadate.Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)
Immunohistochemical staining (Formalin-fixed paraffin-embedded sections) of human cervix cancer. Phospho-JAK2 (Y1007/Y1008) was detected in paraffin-embedded section. Heat mediated antigen retrieval was performed in citrate buffer (pH 6, epitope retrieval solution) for 20 minutes. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 ug/mL JAK2 (phospho Y1007/Y1008) monoclonal antibody, clone IFE-10 overnight at 4°C. Biotinylated goat anti Rabbit IgG antibody was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.Immunocytochemistry
Immunocytochemical staining of Jurkat cells treated with Pervanadate using JAK2 (phospho Y1007/Y1008) monoclonal antibody, clone IFE-10. Phospho-JAK2 (Y1007/Y1008) was detected in immunocytochemical section. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 minutes. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with JAK2 (phospho Y1007/Y1008) monoclonal antibody, clone IFE-10 overnight at 4°C. DyLight488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.Immunoprecipitation
