Applications
Western Blot
Western blot using GLI2 polyclonal antibody (Cat # PAB10051) shows detection of GLI2 protein in rat testes (Lane 1) and human HEK293 (Lane 2) whole cell lysates (arrowhead).
See Ruppert et al for testing conditions.
Each lane contains approximately 35 ug of lysate.
Primary antibody was used at a 1:400 dilution in 5% BLOTTO in PBS overnight at 4°C.
The membrane was washed and reacted with a 1:10,000 dilution of IRDye® 800 conjugated Gt-a-Rabbit IgG [H&L] MX10 for 45 min at room temperature (800 nm channel, green).
Molecular weight estimation was made by comparison to prestained MW markers in lane M (700 nm channel, red).
IRDye® 800 fluorescence image was captured using the Odyssey® Infrared Imaging System developed by LI-COR.

Immunohistochemistry of GLI2 polyclonal antibody (Cat # PAB10051) shows strong cytoplasmic and membranous staining of tumor cells in human breast tissue.
Tissue was formalin-fixed and paraffin embedded.
Brown color indicates presence of protein, blue color shows cell nuclei.

This gene encodes a protein which belongs to the C2H2-type zinc finger protein subclass of the Gli family. Members of this subclass are characterized as transcription factors which bind DNA through zinc finger motifs. These motifs contain conserved H-C links. Gli family zinc finger proteins are mediators of Sonic hedgehog (Shh) signaling and they are implicated as potent oncogenes in the embryonal carcinoma cell. The protein encoded by this gene localizes to the cytoplasm and activates patched Drosophila homolog (PTCH) gene expression. It is also thought to play a role during embryogenesis. The encoded protein is associated with several phenotypes- Greig cephalopolysyndactyly syndrome, Pallister-Hall syndrome, preaxial polydactyly type IV, postaxial polydactyly types A1 and B.
