Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)
Immunohistochemical analysis of paraffin-embedded human ovarian cancer using ILF3 recombinant monoclonal antibody, clone 7H8 (Cat # RAB07469) on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.Immunofluorescence
Immunofluorescent staining of Hela Cells with ILF3 recombinant monoclonal antibody, clone 7H8 (Cat # RAB07469). The cells were fixed in 4% formaldehyde, permeated by 0.2% TritonX-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Rabbit IgG (H+L).Enzyme-linked Immunoabsorbent Assay

This gene encodes a double-stranded RNA (dsRNA) binding protein that complexes with other proteins, dsRNAs, small noncoding RNAs, and mRNAs to regulate gene expression and stabilize mRNAs. This protein was first discovered to be a subunit of the nuclear factor of activated T-cells (NFAT); a transcription factor required for T-cell expression of interleukin 2. NFAT is a heterodimer of 45 kDa and 90 kDa proteins, the larger of which is the product of this gene. These proteins have been shown to affect the redistribution of nuclear mRNA to the cytoplasm. Knockdown of NF45 or NF90 protein retards cell growth; possibly by inhibition of mRNA stabilization. In contrast, an isoform (NF110) of this gene that is predominantly restricted to the nucleus has only minor effects on cell growth when its levels are reduced. Alternative splicing results in multiple transcript variants encoding distinct isoforms
