Applications
Western Blot
Western blot analysis of Hela (A), A549 (B), HL60 (C), U2OS (D), U87MG (E), mouse brain (F), rat brain (G) whole cell lysates with CCT2 recombinant monoclonal antibody (Cat # RAB02580).Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)
Immunohistochemical analysis of human lung cancer formalin fixed paraffin embedded tissue section using CCT2 recombinant monoclonal antibody (Cat # RAB02580). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.93). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.Immunocytochemistry
Immunofluorescence
Immunofluorescent analysis of HeLa cells with CCT2 recombinant monoclonal antibody (Cat # RAB02580). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.Immunoprecipitation

HyperlinkGene Summary
This gene encodes a molecular chaperone that is member of the chaperonin containing TCP1 complex (CCT), also known as the TCP1 ring complex (TRiC). This complex consists of two identical stacked rings, each containing eight different proteins. Unfolded polypeptides enter the central cavity of the complex and are folded in an ATP-dependent manner. The complex folds various proteins, including actin and tubulin. Alternate transcriptional splice variants of the gene described in this record have been observed but have not been thoroughly characterized. [provided by RefSeq
